Fucoidan is a sulfated polysaccharide mainly found in various species of brown seaweeds such as Mozuku, Wakame, Kombu, and Bladderwrack. In other words, Fucoidan is a slime component similar to mozuku and other dark brown seaweeds’ surfaces and is considered a water-soluble dietary fiber. Chemically, it is a polysaccharide that primarily consists of fucose and sulfate.

S. japonica fucoidans were isolated using enzyme-assisted extraction. Enzymatic extracts of S. japonica was prepared using a previously reported method35. S. japonica was washed, dried, and ground, and the brown algal powder was passed through a 40-mesh screen and collected. The powder was dissolved in distilled water in a ratio of 1:30 (w/v). Subsequently, the pH was adjusted to 4.8, and Celluclast and pectinase were added at dry weights of 2.5% and 0.26%, respectively. The reaction solution was stirred at 50 °C for 2 h, and the pH was adjusted to 8.0, followed by the addition of alkaline pectinase (0.3%). The sample was incubated at 60 °C for another 1.5 h. Next, the reaction products were centrifuged, and the supernatants of the S. japonica extracts were collected.
The extracts were mixed with 35% CaCl2 as a percentage of algal dry weight to precipitate alginic acid. Subsequently, the solution was centrifuged, and the collected supernatant was mixed with ethanol to a final ethanol concentration of 70% and stored at 4 °C overnight. The precipitate was collected and re-dissolved with the addition of ethanol at a ratio of 30% and stored at 4 °C. After incubation for 4 h, the solution was centrifuged, and the level of ethanol was modified to 70% and stored at 4 °C overnight. Finally, the fucoidan precipitate was obtained via centrifugal separation and lyophilization.
